Comparative Effects of L-histidine on the Activities of 5'-nucleotidase and Alkaline Phosphatase.

a-Amino acids have been shown to exert a substantial inhibitory effect on the hydrolysis of sodium /3-glycerophosphate by tissue alkaline phosphatases at optimal pH (1). The inhibition by r,-histidine was particularly marked; inhibition constants (Ki) of approximately 6 X 10-S M and 3 X lOma M were obtained for rat intestinal and rat bone phosphatases, respectively (1). The primary concern of this study is to compare the effects of L-histidine on the hydrolysis of adenosine 5’-monophosphate by alkaline phosphatases from bone, intestine, and liver with the effects on 5’-nucleotidase. Particular attention is given to the influence of pH and of Mg* concentration on the existence and magnitude of the effects produced by L-histidine. Data on some other amino acids are also presented.
